Description

Vil1-p2a-FlpO mice express FlpO recombinase from the mouse villin 1 (Vil1) promoter in epithelial cells of the lower gastrointestinal tract.

Detailed description

Vil1-p2a-FlpO mice express FlpO recombinase from the mouse villin 1 (Vil1) promoter in epithelial cells of the lower gastrointestinal tract. VIL1 protein expression has not been examined, but is anticipated to be unaffected by the mutation.

Crosses with an Flp recombinase reporter strain (e.g. Stock No. 021875 or Stock No. 026942) demonstrate expression in epithelial cells throughout the length of the intestine with striking specificity. Reporter expression is not observed in most other tissues examined, including most brain regions, spinal cord, peripheral ganglia, and enteric neurons; rare expression has been noted in taste papillae, epiglottis, pancreas, liver, and thalamus. Intercrosses of Neurod1-Cre (Stock No. 028364) and Vil1-p2a-FlpO mice demonstrate highly selective expression in enteroendocrine cells, with only occasional expression observed in cells of the pancreas.

Heterozygous and homozygous Vil1-p2a-FlpO mice are viable and fertile.

Development

CRISPR/Cas9 methodologies were used to replace the Stop codon of the mouse Vil1 gene with p2A-FlpO recombinase sequence. The mutation was created in C57BL/6J embryos. Resultant mice were backcrossed to C57BL/6J for at least 3 generations by the donating laboratory (see SNP note below).

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve C57BL/6J (Stock No. 000664).

In 2023, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Two of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating, including the rd8 recessive mutation associated with retinal degeneration on Chromosome 1 within the Crb1 locus. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
