Ptk6-/- mice have a neomycin resistance (neo) cassette replacing the transcription start site and the first exon of the PTK6 protein tyrosine kinase 6 (Ptk6) gene, abolishing endogenous gene expression. PTK6 is a cytoplasmic nonreceptor protein kinase involved in the regulation of signaling pathways that control the growth, differentiation, and maintenance of normal epithelial cells as well as tumor cells. It has also been found to be induced in human cancers. PTK6 is highly expressed in gastrointestinal tract with expression also evident in breast tumor cells, and cells of the skin and prostate. Expression is developmentally regulated and is first evident as epithelial linings mature. Homozygous mice are viable and fertile and normal in body weight and length. They exhibit increased villus epithelial cell proliferation and expansion of the proliferative zone, as well as enhanced villus length. They also show a delay in expression of intestinal fatty acid binding protein yet fat absorption was comparable to that seen in wildtype mice.
