Description

Rxfp2^fl/fl floxed mice have loxP sites flanking exon 4 of the Rxfp2 gene making them useful for studying the role of INSL3/RXFP2 signaling during testicular development and descent.

Detailed description

Rxfp2^fl/fl floxed mice have loxP sites flanking exon 4 of the relaxin/insulin-like family peptide receptor 2 (Rxfp2) gene. Rxfp2 encodes a leucine-rich G protein-coupled 7-transmembrane receptor. It is a receptor for the peptide hormone insulin-like 3 (INSL3) which is produced in fetal and adult Leydig cells and is first detected at E13.5 in mouse just before the start of testicular descent. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 4 deleted in the cre-expressing tissues, leading to a reading frame shift and premature stop codon in the resulting Rxfp2^Δ mice.

When bred to mice expressing Cre recombinase in developing embryos, including in the metanephric mesenchyme and gubernaculum, under direction of the retinoic acid receptor, beta (Rarb) promoter, resulting Rxfp2^Δ conditional knock-out males display cryptorchidism with adult testes located in a high intraabdominal position below the kidney.

Development

The L1L2_Bact_P cassette was inserted at position 149966061 of Chromosome 5 upstream of exon 4 of the relaxin/insulin-like family peptide receptor 2 (Rxfp2) gene. The cassette is composed of a frt site followed by lacZ sequence and a loxP site. This first loxP site is followed by neomycin resistance (neo) cassette, a second frt site and a second loxP site. A third loxP site was inserted downstream of exon 4. The mutation was generated in C57BL/6N-derived JM8.N4 embryonic stem (ES) cells. The donating investigator acquired correctly targeted ES cells and injected them into blastocysts. The resulting chimeric mice were bred to ROSA26-Flpe mice (Stock No. 009086) to remove the frt-flanked lacZ and neomycin cassettes. Resulting Rxfp2^fl mice were bred to C57BL/6J mice for at least one generation and mice were subsequently intercrossed to obtain homozygous mice. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
