Teashirt zinc finger family member 1 (encoded by Tshz1) is a transcription factor implicated in olfactory bulb and motor neuron development, craniofacial morphogenesis as well as learning and memory pathways. TSHZ1 is strongly expressed in subtypes of developing and mature neurons of the mouse. The Tshz1-2A-FlpO knock-in mutation has a 2A sequence and an optimized Flp recombinase (FlpO) gene replacing the Tshz1 STOP codon, this is designed to have the endogenous promoter/enhancer elements direct FlpO expression to TSHZ1-positive cells. Tshz1-2A-FlpO homozygous mice are viable and fertile with no overt phenotype. When Tshz1-2A-FlpO mice are bred with mice containing FRT-flanked sequences, FlpO-mediated recombination will result in deletion of the FRT-flanked sequences in Tshz1-expressing cells in the offspring. The donating investigator reports FlpO recombinase expression/activity in Tshz1-expressing cells of the brain, including the olfactory bulb, the piriform cortex, the dorsal and ventral striatum, and the thalamus. The donating investigator did not examine FlpO expression in tissues other than brain. This knock-in allele is designed to avoid disrupting expression of the endogenous protein, although Tshz1 expression from the Tshz1-2A-FlpO allele has not been evaluated. The strain also contains an intact floxed neomycin selection cassette.

Tshz1-2A-FlpO knock-in mice are designed to have optimized Flp recombinase expression directed to teashirt zinc finger family member 1-expressing cells. These mice may be used to generate conditional mutations for studying the function of TSHZ1 in neuronal differentiation, craniofacial development, and complex cognitive pathways.
