Growth factor receptor bound protein 10 (GRB10) is an adapter protein, encoded by an imprinted gene and implicated in embryonic development, cellular growth, insulin signaling, neuronal development and regulation of social behaviors. The Grb10 gene is imprinted in a tissue-specific manner, with paternal expression in a subset of neurons and maternal expression in non-neuronal cells of the brain and in most other adult tissues in the mouse. Heterozygous Grb10P2A-H2B-Venus knock-in mice are viable and fertile. The donating laboratory has not attempted to produce homozygous Grb10P2A-H2B-Venus mice. These knock-in mice have a bicistronic reporter replacing the stop codon of exon 18 of the Grb10 gene. Specifically, this vector includes (from 5' to 3') an in frame replacement of the endogenous Grb10 stop codon with a P2A-H2B-Venus non-gene disruptive reporter cassette. The H2B-Venus nuclear reporter is a fusion protein consisting of the coding sequence of human histone 2B (human HIST1H2BB gene [histone 1 H2bb]) N-terminally fused to the yellow fluorescent protein Venus. Endogenous Grb10 expression is as expected in the brain, with nuclear Venus expression in cells that express the imprinted Grb10 locus. As determined by direct fluorescence, paternal transmission of this Grb10P2A-H2B-Venus knock-in allele yields Grb10 expression throughout the brain and maternal transmission results in Grb10 expression in vasculature-related brain cells and restricted neurons in subcortical areas (Szelenyi E and Osten P 2017, graduate thesis, <a href="https://ir.stonybrook.edu/xmlui/handle/11401/76569">Imprinted Effects in The Brain: From The X Chromosome to Single Autosomal Loci</a>). To date, reporter expression in peripheral tissues has not been assessed. These Grb10P2A-H2B-Venus knock-in mice may be useful in studying Grb10 function and imprinted-expression patterns in cell proliferation, apoptosis, metabolism, neurogenesis and social behavior.
