The Spp1 (secreted phosphoprotein 1, also known as osteopontin) gene encodes a glycoprotein that is a member of the SIBLINGs (Small Integrin Binding LIgand N-Glycosylated proteins) family. It has a variety of functions including a role in inflammation (upregulation of IFNG and IL12), and specifically, intestinal inflammation, as a bone extracellular structural protein with roles in bone remodeling and biomineralization, in apoptosis, and in microglia-synapse phagocytosis. These conditional Spp1fl mice possess loxP sites flanking exons 2 and 3 of the Spp1 gene. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have the exons 2 and 3 deleted in Cre-expressing tissues. Spp1fl mutant mice may be useful in generating conditional mutations to study intestinal epithelial lymphocyte homeostasis (IEL), colitis, and inflammatory bowel disease.
