Description

Sftpc–DreER knock-in mice have a Dre/ERT fusion gene inserted in frame with the translational start codon (ATG) of the surfactant associated protein C (Sftpc) gene. These mice may be used to generate tamoxifen-induced conditional mutations for studying lineage mapping of bronchioalveolar stem cells.

Detailed description

The Sftpc gene encodes pulmonary-associated surfactant protein C which is produced at high levels by type II alveolar epithelial cells in the lung. These Sftpc-DreER knock-in mice carry a Dre/ERT fusion gene in frame with the translational start codon (ATG) of the Sftpc gene. When crossed with a strain containing a rox site flanked sequence of interest, the offspring are useful for generating tamoxifen-induced, Dre-mediated targeted deletions of the rox-flanked sequences specifically in Type II alveolar cells of the lung. Mice homozygous for this Sftpc-DreER knock-in allele are viable and fertile.

Development

The Sftpc-DreER targeting vector was designed to insert cDNA encoding Dre recombinase fused with a mutant form of the estrogen-receptor hormone-binding domain in frame with the translational start codon (ATG) of the surfactant associated protein C (Sftpc) gene. The construct also included a SV40 (polyA) sequence and an frt-flanked PGK-EM7-neomycin resistance cassette. The construct was electroporated into unspecified embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred to 129 mice to confirm germline transmission. Resulting Sftpc-DreER mice were maintained on a mixed B6;129 genetic background by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
