Description

TgH transgenic mice have the mouse insulin I (Ins1) promoter driving mouse microRNA-30d overexpression and EGFP expression in pancreatic beta-cells.

Hemizygous TgH mice have 3 copies of the transgene and 12-fold the level of microRNA-30d expression as the endogenous level in β-cells. Hemizygous TgL mice (Stock No. 037886) are also available. They have 1 copy of the transgene and low expression (2-fold endogenous levels). EGFP expression in both lines is detectable by immunofluorescence.

Detailed description

TgH transgenic mice have the mouse insulin I (Ins1) promoter driving mouse microRNA-30d (Mir30d) overexpression and EGFP expression in pancreatic beta-cells. Dysregulation of microRNA-30d has been reported in the individuals with diabetes. miR-30d is highly enriched in human islets and downregulated in islets isolated from individuals with diabetes and in diabetic db/db mice.

Hemizygous TgH mice have 3 copy of the transgene and have 12-fold the level of microRNA-30d expression as the endogenous level in β-cells, with no expression differences seen in brain, liver, heart, spleen or kidney. GFP reporter expression, as detected by immunostaining, is restricted to 50-60% of insulin-producing β-cells. TgH mice display significant diet-induced hyperglycemia and β-cell dysfunction. They exhibit downregulation of β-cell-enriched genes and upregulation of α-cell enriched genes. In addition, loss of β-cell identity is accompanied with increased insulin/glucagon-double positive bihormonal cells and excess plasma glucagon levels.

The donating investigator suggests breeding hemizygous mice to C57BL/6J inbred mice (Stock No. 000664) or wildtype littermates to maintain the copy number.

TgL (Stock No. 037886) is a companion strain to this line. TgL mice have low expression (2-fold endogenous levels) and 1 copy of the transgene. EGFP expression in both lines is detectable by immunofluorescence.

Development

Transgenic miR-30d mice were created to express murine microRNA-30d (Mir30d) and EGFP under control of the mouse insulin I (Ins1) promoter. The linearized transgene was purified and then microinjected into the fertilized eggs of mixed C57BL/6 and SJL mice. These TgH mice, with high transgene expression (12-fold) and 3 copies of the transgene, were selected and bred to C57BL/6J mice (Stock No. 000664) for at least 20 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
