Tissue-specific expression of the mouse Kcnab1 (potassium voltage-gated channel, shaker-related subfamily, beta member 1) can be regulated with the administration of doxycycline after breeding a mouse carrying this transgene with another transgenic mouse expressing reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA).

Detailed description

TRE-Kvβ1.1 transgenic mice express the mouse Kcnab1 (potassium voltage-gated channel, shaker-related subfamily, beta member 1 or Kvβ1.1) under the control of a tetracycline-responsive promoter element (TRE; tetO). Kcnab1 encodes a member of aldo-ketoreductase superfamily that binds NAD(P)(H) and is involved in the regulation of voltage-gated potassium currents (Kv). Kvβ proteins are expressed in the coronary vasculature, in contrast to Kvβ2, Kvβ1.1 functions to inhibit Kv-mediated oxygen-dependent coronary vasodilation. Hemizygotes are viable and fertile with no overt phenotype. The donating investigator did not attempt to generate homozygotes. When hemizygotes are bred with another transgenic mouse expressing reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA) to create bitransgenic animals, tissue-specific Kvβ1.1 transgene expression can be regulated with the tetracycline analog, doxycycline.

For example, when combined with mice expressing tTA under the control of a smooth muscle promoter (Tg(Tagln-rtTA)E1Jwst; Stock No. 006875), bitransgenic animals treated with dox exhibit increased Kv1.5:Kvβ1-associated proximity ligation signal in coronary arterial myocytes and blunted vasodilation of isolated mesenteric arteries.

Development

The transgene was designed with a sequence encoding mouse Kcnab1 (potassium voltage-gated channel, shaker-related subfamily, beta member 1) under the control of the Tet-responsive element promoter (TRE; 2^nd generation) and was injected into fertilized FVB/N mouse eggs. Founder line 1 was subsequently established and crossed to FVB/N mice to expand the line by the donating investigator (see SNP note below). Upon arrival at The Jackson Laboratory, the mice were crossed to FVB/NJ (Stock No. 001800) at least once to establish the colony.

A 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. One of the 43 markers on Chromosome 14 was segregating with an unknown source.
