iBE (in vivo/inducible Base Editing) knock-in mice were created using recombinase-mediated cassette exchange site downstream of the Col1a1 gene. iBE mice express a cytosine base editor (C:G to T:A) under the control of a tetO promoter (TRE), which allows inducible and transient expression. When combined with a strain expressing tetracycline-controlled transactivator protein (tTA) or reverse tetracycline-controlled transactivator protein (rtTA), tissue-specific iBE expression in the resulting double positive offspring can be regulated with tetracycline or its analog doxycycline (dox). Induction of the base editor in the presence of a specific sgRNA enables targeted base editing across a broad range of tissues with no evidence of DNA or RNA off-target effects. Mice homozygous for the mutation are viable and fertile. The iBE strain may be used to create individual or multiple single nucleotide variants (SNVs) in pancreatic, lung, and intestinal organoids and somatic cells for testing in pre-clinical cancer models.
