Description

Pi*M mice express human SERPINA1 (also known as AAT or α1-antitrypsin), which carries the common M variant (PI*M). PI*M mouse may be used a coisogenic control for the PI*Z transgenic mouse (Stock No. 037670), which carries a glutamic acid to lysine subsitution at residue 342 (E342K). The PI*Z variant is associated with alpha-1 antitrypsin (AAT) deficiency.

Detailed description

SERPINA1 (serine (or cysteine) peptidase inhibitor, clade A, member 1 cluster, also known as AAT or α1-antitrypsin) encodes an abundant serine protease inhibitor expressed primarily in the liver. There are at least 30 variants of AAT found in humans, the most common being the M variant or PI*M, which carries a glutamic acid at residue 342. PI*M mice express human SERPINA1 in the liver as well as in polymorphonuclear leukocytes in addition to their endogenous AAT. PI*M transgenic mice are viable and fertile. Response to LPS treatment in transgenic PI*M mice is similar to wild-type and does not alter the unfolded protein response (UPR) signaling branches. The PI*M mouse may be used a coisogenic control for the PI*Z transgenic mouse (Stock No. 037670), which carries a glutamic acid to lysine substitution at residue 342 (E342K). The PI*Z variant is associated with alpha-1 antitrypsin (AAT) deficiency.

Development

The transgenic construct contains the entire human SERPINA1 (serine (or cysteine) peptidase inhibitor, clade A, member 1 cluster) gene under its endogenous promoters (both hepatocyte and macrophage) plus 5 kB of the 5' and 3 kb of the 3' flanking genomic DNA sequences. The linearized construct was microinjected into fertilized C57BL/J oocytes. Founder 1 carrying 4 copies of the transgene was maintained on the C57BL/6J background. Upon arrival, mice were bred to C57BL/6J for at least 1 generation to establish the colony.
