Product name: Pogz^f
Research areas: Hematological Research, Research Tools

Description

Pogz^f mice possess loxP sites flanking exons 13-19 of the pogo transposable element with ZNF domain (Pogz) gene. These floxed mice are useful for tissue specific deletion of Pogz when studying hemoglobin switching and β-globin disorders.

Of note, the donating investigator has made mice available with the following alleles: Id2^fl (Stock No. 037663), Id2^eYFP knock-in/knock-out (Stock No. 037664), Pogz^f (Stock No. 037665) and Id1^fl (Stock No. 037666).

Detailed description

Pogz (pogo transposable element with ZNF domain) encodes a zinc finger protein with a transposase domain involved in transcriptional regulation. POGZ is expressed in hematopoietic progenitor cells and is involved in hemoglobin switching.

Pogz^f mice possess loxP sites flanking exons 13-19 of the Pogz gene. Exons 13-19 encode three zinc fingers, the CENPB domain, and the DDE domain. Mice homozygous for the allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 13-19 deleted in the cre-expressing tissues. Germline deletion of Pogz results in embryonic lethality or death shortly after birth.

Bone marrow transfer experiments using bone marrow cells extracted from Pogz^f mice crossed to mice expressing an inducible Cre recombinase (Tg(Mx1-cre)1Cgn (Stock No. 003556)), transplanted into irradiated mice treated with pIpC demonstrate increased Hbb-y expression in peripheral blood cells, suggesting that Pogz is involved in the negative regulation of embryonic β-globin (Hbb-y) in adult red blood cells.

Development

The Pogz^f targeting vector was designed to insert a loxP site upstream of exon 13, and an FRT-flanked neomycin cassette followed by a loxP site downstream of exon 19 (a 4.13 kb region) of the pogo transposable element with ZNF domain (Pogz) gene on chromosome 3. Exons 13-19 encode three zinc fingers, the CENPB domain, the DDE domain and part of the 3' untranslated region. The construct was electroporated into 129S1/Sv-derived CJ7 embryonic stem (ES) cells. Resulting ES cells were injected into blastocysts and resulting mice were bred to ACTB:FLPe mice (Stock No. 005703) to remove the neo cassette. Progeny were crossed to remove the Flp-expressing transgene, and offspring were bred to C57BL/6Ncr mice for at least 10 generations prior to sending males to The Jackson Laboratory Repository. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Symbol: Pogz^tm1.1Jrk
Name: targeted mutation 1.1, Jonathan R Keller
MGI accession ID: MGI:6360951
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed); No functional change
Synonyms: Pogzf
Marker symbol: Pogz
Marker name: pogo transposable element with ZNF domain
Marker synonyms: 9530006B08Rik; MRD37; WHSUS; ZNF280E; ZNF635; ZNF635m
Chromosome: 3
Strain of origin: 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺
Molecular note: A loxP site was inserted upstream of exon 13. An FRT-flanked neomycin resistance cassette with a 3' loxP site was inserted downstream of exon 19. Flp-mediated recombination removed the selection cassette and left exons 13 through 19 floxed.
