Description

This tamoxifen-inducible pdx1^PB CreER^TM strain on the diabetes susceptible NOD background may be used to create inducible pancreatic cell knockouts when crossed with strains carrying floxed alleles.

Detailed description

This inducible transgenic strain pdx1^PB CreER^TM expresses Cre/Esr1 from the mouse pancreatic endocrine-specific Pdx1 enhancer and hsp68 minimal promoter on the diabetes susceptible NOD background. Cytoplasmic stores of Cre recombinase translocate to the nuclear compartment upon induction with tamoxifen. When crossed with a strain containing a loxP-flanked sequence of interest, the offspring are useful for generating tamoxifen-induced, cre-mediated targeted deletions. Cre expression can be detected as early as E14.5 following tamoxifen injection at E12.5. Mice hemizygous for the mutation are viable and fertile. Spontaneous autoimmune diabetes develops in a similar time frame to standard NOD mice.

pdx1^PB CreER^TM mice crossed with R26R floxed lacZ reporter mice (see Stock No. 003474) and then injected with tamoxifen express lacZ in 86% of β cells, 75% of α cells and 85% of γ cells at E16.5 with expression maintained in 90% of β cells throughout adulthood. The donating investigator indicates that there is slight cre leakiness (<1%) in the pancreas in the absence of tamoxifen.

Development

The pdx1^PB CreER^TM transgene was designed by Dr. Maureen Gannon (Vanderbilt University) to have a 1 kb mouse Pdx1 pancreatic islet specific enhancer and an hsp68 minimal promoter driving expression of CreER^TM fusion gene (Cre recombinase fused to a G525R mutant form of the mouse estrogen receptor ligand binding domain) cDNA followed by polyA sequences. The transgene was introduced into (C57BL/6 x DBA/2) donor eggs. Founder line 1, which exhibits the highest expression, was established on a mixed background. Mice were subsequently backcrossed onto the NOD/ShiLtJ (Stock No. 001976) background by Dr. Raghavendra Mirmira (University of Chicago) for more than 10 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize NOD/ShiLtJ oocytes (Stock No. 001976).
