Description

Mogat1^loxP mice carry a knock-in allele of the mouse Mogat1 (monoacylglycerol O-acyltransferase 1; also called MGAT1) gene in which exon 4 is flanked by loxP sites. Cre recombinase-mediated excision of the floxed region results in a knock-out alleleallele that has been useful in studies of adipocyte and hepatocyte metabolism.

Detailed description

Mogat1^loxP mice carry a knock-in allele to the mouse Mogat1 (monoacylglycerol O-acyltransferase 1; also called MGAT1) gene in which exon 4 is flanked by loxP sites. Cre recombinase-mediated excision of the floxed region results in a knock-out allele useful in studies of diacylglyceride and triglyceride metabolism. Mogat1 is expressed in adipose, liver, intestine, and stomach.

Knock-out of MOGAT1 in adipocytes (through crosses with Stock No. 028020) leads to higher rates of basal adipocyte lipolysis. The gene may play a role in regulating basal adipocyte free fatty acid (FFA) retention during lipolysis of triglycerides. Knock-out of MOGAT1 in liver (through crosses with Stock No. 003574) does not lead to any discernible differences between groups.

Development

An FRT-lacZ-loxP-neomycin-FRT-loxP cassette was introduced to intron 3 and a loxP site was placed in intron 4 via homologous recombination in C57BL/6N-derived JM8A.N4 embryonic stem (ES) cells (MMRRC: 058725-UCD; CSD35789). Resultant mice were crossed with C57BL/6 background mice widely expressing Flp recombinase (Stock No. 005703) to remove the floxed lacZ/neomycin cDNA cassette, leaving exon 4 flanked by loxP sites.

Upon arrival at The Jackson Laboratory, sperm was cryopreserved. Cryorecovery/live colony development will involve C57BL/6NJ (Stock No. 005304). (July, 2022)

In 2022, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. All 5 markers that determine C57BL/6J from C57BL/6N were found to be derived from C57BL/6J.

Molecular note: The L1L2_Bact_P cassette was inserted at position 78499742 of Chromosome 1 upstream of the critical exon(s) (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 78500675. The critical exon(s) is/are thus flanked by loxP sites.A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele to remove the lacZ sequence and neo selection cassette, leaving loxP sites flanking the critical exon(s).
