Description

The granzyme-B-Cre transgene is designed to have the human GZMB (granzyme B) promoter directing Cre recombinase expression to activated T cells.

In addition to this NOD/ShiLtJ congenic background granzyme-B-Cre transgenic strain (Stock No. 037455), the granzyme-B-Cre transgene is also available on a B6;FVB mixed genetic background (Stock No. 003734).

Detailed description

The granzyme-B-Cre transgene is designed to have the human GZMB (granzyme B) promoter directing cre recombinase expression to activated T cells. When granzyme-B-Cre mice are bred with mice containing loxP-flanked sequences, cre-mediated recombination will result in deletion of the floxed sequences in GZMB-expressing cells in the offspring.

Mice homozygous for the granzyme-B-Cre are viable and fertile, however homozygotes may exhibit an increased aggression phenotype.

This granzyme-B-Cre transgenic strain is one component of a cre-recombinase mediated mouse model system that allows irreversible labeling of antigen-stimulated T cells with surface expressed ALPP (placental alkaline phosphatase).

Although it has not been determined if these mice develop autoimmune type 1 diabetes, the donating investigator suggests a footpad injection of Complete Freund's Adjuvant (CFA) administered once at weaning to delay diabetes onset, thus extending the lifespan of breeders.

In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.

This allele is also available on a B6;FVB genetic background as Stock No. 003734.

Development

A targeting construct containing the human granzyme B promoter, the SV40 nuclear localization signal sequence, cre recombinase, and the human growth hormone polyadenylation sequence was injected into the pronuclei of fertilized FVB/N eggs. Founder mice were obtained and bred. Founder animals were backcrossed to C57BL/6 and are available on a mixed genetic background as (Stock No. 003734). Dr. Yi-Guang Chen (Medical College of Wisconsin) obtained some of these animals and further backcrossed them to NOD/ShiLtJ mice (Stock No. 001976) for at least 11 generations before sending males back to The Jackson Laboratory. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize NOD/ShiLtJ oocytes.
