Description

M1Red transgenic mice express a DsRed2 reporter directed by mouse Irgm1 (immunity-related GTPase family M member 1) promoter elements. DsRed2 activity is observed following interferon stimulation. This strain may be used to visualize cytokine expression, specifically interferon signaling in vivo.

Detailed description

M1Red transgenic mice express the DsRed2 fluorescent protein under the control of the mouse Irgm1 (immunity-related GTPase family M member 1) promoter.  Irgm1 is a GTPase with wide expression in cells of the hematopoietic and non-hematopoietic lineages; it is upregulated by induction by different types of interferons in a STAT1-dependent manner. Fluorescence is observed following inoculation with individual recombinant interferons or influenza virus A. Mice homozygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These mice may be used to visualize cytokine expression, specifically interferon signaling in vivo.

Development

The bacterial artificial chromosome clone, RP23-305A21, containing the mouse Irgm1 gene was modified to insert a DsRed2 reporter into the Irgm1 ATG translational start site using the galK replacement method. Following sequence verification, the modified BAC was microinjected into the pronuclei of fertilized C57BL/6N oocytes. Founder line 1 was bred to C57BL/6NCrl mice. The donating investigator indicates that no other genes are expressed from the BAC. The copy number and transgene insertion site are unknown. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

In 2022, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. One of the 5 markers that determine C57BL/6J from C57BL/6N was found to be segregating on Chromosome 6. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
