Description

OMP-H2B::Cherry knock-in/knock-out mice may be useful for visualization of mature olfactory sensory neurons.

Detailed description

OMP-H2B::Cherry knock-in/knock-out mice have an H2B::mCherry fusion protein replacing the entire coding region of the olfactory marker protein (Omp) gene. Under direction of the murine Omp promoter, these mice express nuclear mCherry in the intermediate layer of the olfactory epithelium. Homozygous mice are viable and fertile.

mCherry fluorescence is detectable by FACS, immunohistochemistry, and direct fluorescence.

Development

The OMP-H2B::Cherry targeting vector, created by Dr. Paul Feinstein (Hunter College), contains an H2B::mCherry fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] with a C-terminal red fluorescent protein gene) followed by polyadenylation (pA) sequence and a PGK-puromycin resistance (puro)-pA cassette and was used to replace the entire coding region of the olfactory marker protein (Omp) gene. The construct was electroporated into B6;129 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting OMP-H2B::Cherry mice were crossed to B6129SF1/J mice (Stock No. 101043) for at least 10 generations by the donating laboratory. Mice were maintained on a mixed B6;129 genetic background. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
