Description

Mov10^flox mice possess loxP sites flanking exons 6-7 of the Mov10 gene. These mice may be useful when studying the role of MOV10 in neurogenesis and brain function, such as learning and memory.

Detailed description

Mov10 RISC complex RNA helicase (MOV10) preferentially binds RNAs involved in early neuronal development and brain function. These Mov10^flox mice possess loxP sites flanking exons 6-7 of the Mov10 gene. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 6-7 deleted in the cre-expressing tissues, leading to a reduction of MOV10 protein. The donating laboratory bred these Mov10^flox mice to an Emx1^Cre mouse line (Stock No. 005628) to reduce MOV10 in excitatory neurons of the neocortex and hippocampus. The resulting conditional knock-out mouse (Mov10 cKO) had a ~50% reduction in MOV10 expression in brain. To obtain a greater reduction, the lab crossed Mov10 cKO to a Mov10 heterozygote line (described in Skariah et al., 2017), which resulted in the MOV10 deletion mouse that has a 90% reduction of MOV10 protein in postnatal day 0 (P0) brain tissue. These brain-specific Mov10 KO mice showed enhanced fear memory, increased cortical thickness and increased dendritic arborization of hippocampal neurons compared to controls. Mov10^flox mice may be useful when studying neuronal development and brain function, such as learning and memory.

Development

The Mov10^flox allele was generated using a Mov10^{tm456285a(L1L2_GT2_LF2A_LacZ_BetactP_neo)} construct obtained from the European Mouse Mutant Cell Repository (EMMCR). The targeting cassette is composed of an FRT site followed by an EN2 splice acceptor site, a lacZ sequence, a polyadenylation (polyA) site, an initial loxP site followed by a neomycin (neo) cassette, a second FRT site, and a second loxP upstream of exon 6 of the Mov10 RISC complex RNA helicase (Mov10) gene. A third loxP site is inserted downstream of exon 7 of the Mov10 gene. The construct was electroporated into C57BL/6N-derived embryonic stem (ES) cells. Targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were bred with B6.129S4-Gt(ROSA)26Sor^tm1(FLP1)Dym/RainJ (Stock No. 009086) mice to excise the FRT-flanked lacZ sequence and neo selection cassette. Mov10^flox mice, with neo cassette removed and floxed exons 6-7 intact, were maintained on a C57BL/6 background by the donating laboratory. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

In 2025, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. All 5 markers that determine C57BL/6J from C57BL/6N were found to be C57BL/6J-like. These data suggest the mice sent to The Jackson Laboratory Repository were on a predominantly C57BL/6J genetic background. It was also determined that the donor males were harboring FLP and Cre alleles; these were removed through selective breeding to C57BL/6J inbred mice for 1-2 generations by The Jackson Laboratory. New males, that were negative for the unwanted FLP and Cre alleles, were then used as donors to generate cryopreserved material .
