Detailed description

Podocan^-/- mice have a neo cassette replacing exons 4-8 of the podocan (Podn) gene, abolishing endogenous gene expression. Podocan is a small leucine-rich repeat protein that is expressed in vascular endothelial cells and vascular smooth muscle cells where it negatively regulates cell proliferation and cell migration. It is also involved in cancer progression, arteriosclerosis, kidney function, and diabetic nephropathy. Homozygous mice are viable and fertile. They have increased arterial lesion formation and smooth muscle proliferation in response to injury.

Development

The podocan^- targeting vector was designed to replace exons 4-8 of the podocan (Podn) gene with a neomycin resistance (neo) cassette. The construct was electroporated into SVJ129-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and the resulting chimeric males were bred to C57BL/6J females. These mice were backcrossed for at least 9 generations to C57BL/6J mice. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
