Tlr2 flx floxed mice have loxP sites flanking exon 3 of the Tlr2 gene. These mice may be useful when studying pathogen recognition and activation of innate immunity.

Detailed description

Tlr2 flx floxed mice have loxP sites flanking exon 3 of the toll-like receptor 2 (Tlr2) gene. TLR2 is a cell-surface receptor which plays a fundamental role in pathogen recognition and activation of innate immunity. This gene has been implicated in the pathogenesis of several autoimmune diseases including inflammatory bowel disease and Crohn’s Disease, as well as some intestinal cancers. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 removed in the Cre-expressing tissues of the offspring.

Development

The Tlr2 flx targeting vector was designed to insert a loxP- and frt-flanked neomycin resistance cassette upstream of exon 3 of the toll-like receptor 2 (Tlr2) gene, and a third loxP site was inserted downstream of exon 3. The construct was electroporated into iTL HF4 (129/SvEv x C57BL/6 FLP)-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and resulting chimeric mice were bred with C57BL/6 mice to generate mice with a germline deletion of the neo cassette. Resulting Tlr2 flx mice were subsequently backcrossed to C57BL/6J mice for at least 10 generations by the donating laboratory (see SNP note below). Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

In 2022, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Three of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
