Description

Ier3-flox conditional mutant mice possess loxP sites flanking exons 1 and 2 of the Ier3 (immediate early response 3) gene and may be useful in generating conditional mutations to study colitis and colitis associated carcinogenesis.

Detailed description

IER3 (immediate early response 3) functions as both a ubiquitous early response gene and a stress-inducible gene that is involved in cell proliferation and survival. Ier3 is induced in various cells by irradiation, viral infection, inflammatory cytokines, chemical carcinogens, increased levels of reactive oxygen species, and apoptotic stimuli. The conditional Ier3-flox mice possess loxP sites flanking exons 1 and 2. Exons 1 and 2 constitute the entire coding region (480 bp) of the gene. Mice homozygous for the mutation are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 1 and 2 deleted in cre-expressing tissues. Deletion of exons 1 and 2 is expected to result in a null allele that abolishes Ier3 expression in a tissue-specific manner.

For example, when bred to a strain expressing Cre recombinase in intestinal epithelial cells or mature myeloid cells (such as Tg(Vil1-cre)997Gum, Stock No. 004586, or Lyz2^tm1(cre)Ifo, Stock No. 004781, respectively) progeny treated with DSS (to induce acute colitis) bred to homozygosity exhibit less weight loss and increased survival as compared to wild-type.

Development

The Ier3-flox targeting construct contains a loxP site and an FRT-flanked neomycin cassette upstream of exon 1 and a second loxP site downstream of exon 2 in the mouse Ier3 locus on chromosome 17. The construct was electroporated into unspecified C57BL/6 derived embryonic stem (ES) cells (GenOway). Correctly targeted ES cells were injected into blastocysts. Chimeric males were bred to female mice expressing FLP recombinase on the C57BL/6 background (GenOway) to excise the FRT-flanked neo cassette. The strain was backcrossed to C57BL/6J for at least 3 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

Allele

Symbol: Ier3^tm1.1Hscha
Name: targeted mutation 1.1, Heiner Schaefer
MGI accession ID: MGI:6849742
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed)
Synonyms: Ier3-flox
Marker symbol: Ier3
Marker name: immediate early response 3
Marker synonyms: cAMP inducible gene 3; cI-3; Dif2; DIF-2; gly96; IEX1; IEX-1; IEX-1L; Ler3; Prg1
Chromosome: 17
Strain of origin: C57BL/6
Molecular note: The targeting construct contains a loxP site and an FRT-flanked neomycin cassette upstream of exon 1 and a second loxP site downstream of exon 2. Flp-mediated recombination removed the neomycin cassette.
General note: ES cells = C57BL/6 GenOway
