BAC-Sprr2f-Cre transgenic mice express Cre recombinase under the control of the small proline-rich protein 2F (Sprr2f) promoter. Sprr2f encodes a protein indirectly involved in the non-cell autonomous inhibitory effect on fibroblast growth factor two signaling. Endogenous gene expression is only seen in the uterus, specifically in endometrial epithelium. Hemizygous mice are viable and fertile, it is unknown if homozygotes are viable. When crossed with a strain containing a loxP site-flanked sequences, Cre-mediated recombination occurs only in the endometrial epithelium. Cre activity begins post-natally, around 5 weeks of age, permitting normal uterine development in females. The donating investigator states that the estrogen dependence of the Sprr2f locus, which harbors estrogen response elements, is retained and that Cre-mediated recombination can be induced early by &beta;-estradiol administration in sexually immature mice at 3 weeks of age. When BAC-Sprr2f-Cre mice are bred to R26R lacZ reporter mice (Stock No. <a href="https://www.jax.org/strain/003474">003474</a>), endometrial epithelium in resulting BAC-Sprr2f-Cre;R26R mice exhibit &sim;50% recombination by 6 wk of age. This recombination efficiency (i.e., subtotal, mosaic) is useful for genetically engineered mouse models of endometrial carcinoma (EC), as it recapitulates the admixture of genetically normal and abnormal epithelial cells that characterize the initial stages of endometrial carcinogenesis. However, the mosaic pattern may not be desirable for functional investigations of factors involved in endometrial biology. When BAC-Sprr2f-Cre transgenic mice are bred to Fbw7flox mice (Stock No. <a href="https://www.jax.org/strain/017563">017563</a>) and Ptenflox mice (Stock No. <a href="https://www.jax.org/strain/006440">006440</a>), resulting offspring develop well-differentiated endometrioid adenocarcinomas, which all eventually develop into definitive uterine carcinosarcomas. Activation of a conditional LSL-PoleP286R/+ allele (see Stock No. <a href="https://www.jax.org/strain/037051">037051</a>) in the endometrium by crosses with these mice is sufficient to elicit endometrial cancers in all animals that closely resembles human POLE ultramutant endometrial cancers, including very high mutational burden. Of note, the donating investigator compared this new BAC transgenic line with his other Cre expressing mouse line, Tg(Sprr2f-cre)1Dcas, which has ectopic expression in cerebellum and kidney. The discrepancy in expression is likely due to the smaller promoter and lack of more distant regulatory elements in the Tg(Sprr2f-cre)1Dcas line.
