Gm5127 is a predicted mouse gene that has no corresponding gene in other species. It is expressed in endothelial cells of the brain, retina, dermis, and colon; expression is not observed in other organs tested (small intestine, diaphragm, bone, kidney, heart, lung, and liver). In embryonic tissues (E9.5 and E13.5), Gm5127 is mainly expressed in non-endothelial cells. In the brain, Gm5127 is restricted to venous endothelial cells and, in the colon, expression is mainly in the capillary endothelium. Gm5127(BAC)CreERT2 transgenic mice contain a mouse Gm5127 promoter directing tamoxifen-inducible Cre recombinase expression. When transgenic mice are bred with mice containing loxP-flanked (floxed) sequences, tamoxifen-inducible cre-mediated recombination will result in deletion of floxed sequences in Gm5127-expressing cells in the offspring. Hemizygous and homozygous mice are viable and fertile with no reported phenotypic abnormalities. This strain may be useful in of angiogenesis and, specifically, in fate mapping of venous endothelial cells in the brain and retinal and dermal vascular in postnatal and adult mice stages.The Cre-ERT2 fusion protein consists of Cre recombinase fused to a triple mutant form of the human estrogen receptor which does not bind its natural ligand (17&beta;-estradiol) at physiological concentrations but will bind the synthetic estrogen receptor ligands 4-hydroxytamoxifen (OHT or tamoxifen) and, with lesser sensitivity, ICI 182780. Restricted to the cytoplasm, Cre-ERT2 can only gain access to the nuclear compartment after exposure to tamoxifen. To counteract the mixed estrogen agonist effects of tamoxifen injections, which can result in late fetal abortions in pregnant mice, progesterone may be coadministered.

Gm5127(BAC)CreERT2 transgenic mice express a tamoxifen inducible Cre recombinase/ERT2 fusion under the control of the mouse Gm5127 promoter elements. Cre recombinase activity is observed specifically in venous endothelial cells. This strain may be useful in studies of angiogenesis, and specifically, in fate mapping of venous endothelial cells in the brain and retinal and dermal vascular in postnatal and adult mice stages.
