Description

Klrc3^-/- mice lack exons 3-5 of the Klrc3 gene. These mice are useful when studying NK cell killing of virus-infected cells.

Detailed description

Klrc3^-/- mice lack exons 3-5 of the killer cell lectin-like receptor subfamily C, member 3 (Klrc3) gene. Klrc3, also known as NKG2E, encodes an activating receptor on natural killer cells, which are lymphocytes that can mediate lysis of certain tumor cells and virus-infected cells without previous activation. CD94 forms inhibitory receptors with NKG2A and activating receptors with NKG2E or NKG2C. Mice that are homozygous for this allele are viable and fertile. Klrc3^-/- mice have no obvious phenotype. A few mice succumb to lethal mousepox 10-11 days post infection, but the majority are resistant, and most of these mice have virus titers in their spleens similar to that of wildtype C57BL/6 mice. NKG2C deficiency does not increase susceptibility to lethal ECTV infection.

Development

The L1L2_Bact_P cassette was inserted at a position on Chromosome 6 downstream of exon 2 of the killer cell lectin-like receptor subfamily C, member 3 (Klrc3) gene, creating a 3581 bp deletion of exons 3-5. The cassette is composed of a FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. The mutation was generated in C57BL/6N-A^tm1Brd-derived JM8A3.N1 embryonic stem (ES) cells. Some of these ES cells were acquired by Dr. Luis J. Sigal (Thomas Jefferson University) and were injected into blastocysts. The resulting chimeric mice were bred to C57BL/6NCrl mice, and an F1 carrier was bred to ACTB:FLPe mice (Stock No. 003800) to remove the lacZ/neo cassette and then subsequently mice were bred together to remove the FLPe expressing transgene. These Klrc3^-/- mice were bred to and maintained on a C57BL/6NCrl background. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to C57BL/6NJ oocytes (Stock No. 005304).
