Description

Exon 1 of the mouse Bard1 (BRCA1 associated RING domain 1) gene, encoding the initiator methionine and part of the RING domain, is flanked by loxP sites in these Bard1^co conditional mutant mice (Billing, 2018). Cre recombinase-mediated excision of the floxed region results in a knock-out allele, useful in studies of tumor suppression.

Detailed description

Exon 1 of the mouse Bard1 (BRCA1 associated RING domain 1) gene, encoding the initiator methionine and part of the RING domain, is flanked by loxP sites in these Bard1^co conditional mutant mice. Cre recombinase-mediated excision of the floxed region results in a knock-out allele, useful in studies of tumor suppression.

Widespread, homozygous knock-out of the gene (created through crosses with EIIa-cre (Stock No. 003724) mice) is embryonic lethal (Billing, 2018), while mammary epithelial cell-specific inactivation of the gene (created through crosses with Wap^tm1(cre)Arge mice) induces mammary tumors that resemble the basal-like triple-negative breast cancers of women who carry human BRCA1 mutations (Shakya, 2008).

 

Development

A loxP site was introduced ~900 bp upstream of exon 1 (encoding the initiator methionine and part of the RING domain) at an Xmn1 restriction site, and a loxP-FRT-PGK-neo-FRT cassette was introduced ~350 bp downstream of exon 1 via homologous recombination in (C57BL/6J x 129S6/SvEvTac)F1-derived KV1 embryonic stem (ES) cells. The FRT-PGK-neo-FRT cassette was excised through crosses with Tg(ACTFLPe)9205Dym mice (Stock No. 005703), leaving exon 1 flanked by loxP sites (Billing, 2018). This strain was maintained on a mixed C57BL/6-129 genetic background by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
