Description

B10.BR-HA104 (Tg(SV40-HA)HA104Ajca) mice express a full length influenza virus PR8 hemagglutinin (HA) membrane bound neo-self antigen on the B10.BR-H2^k2 H2-T18^a background. This mouse strain may be useful for studies of antigen-specific B cell responses in models of hematopoietic cell transplantation and graft versus host disease.

Detailed description

B10.BR-HA104 (Tg(SV40-HA)HA104Ajca) mice express a full length PR8 hemagglutinin (HA) membrane bound neo-self antigen under the direction of the SV40 promoter. HA contains S1, the major I-Ed-restricted PR8 HA T cell determinant. In this strain, the HA-104 transgene has been introgressed into the H2 variant, B10.BR-H2^k2 H2-T18^a/SgSnJJrep. The H2^k2 haplotype differs from the prototype H2^k in having the a, rather than the canonical b, alleles of the Class Ib genes T18 (formerly Tla) and Qa-1. The H2^k2 haplotype is shared by the inbred strains C57BR/cdJ and C58/J. The allele H2-T18 determines the presence of the antigenic specificities TL.1, 2, 3, 5, 6, 7 and occurs in strains A, C57BR, C58, SJL, and others. Mice hemizygous for the transgene are viable and fertile with no observable phenotype. The viability/fertility of homozygotes has not been assessed. This mouse strain may be useful for studies of antigen-specific B cell responses in models of hematopoietic cell transplantation and graft versus host disease.

On the BALB background, immunized (using intact influenza virus) HA-104 mice develop a subset of HA-specific IgG-secreting (but not IgM) B cells that are substantially reduced in number. However, although primary response B cells are affected by negative selection, an autoreactive memory B cell response develops unimpaired. When combined with Tg(Tcra/Tcrb)1Vbo/Ajca transgenic mice (MMRRC Stock No. 043834) expressing an S1-specific T cell receptor, the population of 6.5^hi CD4+ single positive thymocytes is reduced, however, the percentage of Foxp3+ regulatory T cells is increased within this subset of CD4+ thymocytes

In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.

Development

The HA104 transgenic construct created in the laboratory of Dr. Andrew Caton (Wistar Institute) contains the entire influenza virus A/Puerto Rico/8/34 H1N1(PR8) hemagglutinin (HA) under direction of the SV40 early region promoter/enhancer. Founder line HA104 expresses low levels of transgene mRNA in several tissues, including intestine, thymus, bone marrow, and spleen. The linearized construct was microinjected into fertilized BALB/c x C57BL/6 oocytes. HA104 mice were bred to BALB/cAnNCrl mice for at least 13 generations. Dr. Vedra Radojcic (University of Utah) obtained H104 from the MMRRC at The Jackson Laboratory as MMRRC Stock No. 043831 and introgressed the allele into B10.BR-H2^k2 H2-T18^a/SgSnJJrep (Stock No. 004804) for nine generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize B10.BR-H2^k2 H2-T18^a/SgSnJJrep oocytes.
