Description

Il-10^FL mutant mice possess loxP sites flanking exon 1 of the Il10 (interleukin 10) gene and may be useful in generating conditional mutations to study immunoregulation and inflammation.

Detailed description

Il10 (interleukin 10) encodes a class 2, anti-inflammatory cytokine involved in immunoregulation and inflammation. IL10 is produced primarily by monocytes and is associated with several autoimmune diseases. These conditional Il-10^FL mice possess loxP sites flanking exon 1. Mice homozygous for the mutation are viable and fertile with no observable phenotype. Deletion of exon 1 abolishes Il10 function.

Development

The IL-10^FL mutation was created by Dr. Axel Roers in the laboratory of Dr. Werner Muller at the University of Cologne. The targeting vector was designed to place a loxP site upstream of exon 1 and a loxP-flanked neomycin cassette downstream of exon 1 in the Il10 locus on chromosome 1. The construct was electroporated into 129P2/OlaHsd-derived IB10 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient CB20 blastocysts and chimeric males were bred with C57BL/6 females to establish the mutant colony on a mixed B6;129P2 genetic background. Mice were then crossed to mice B6.C-Tg(CMV-cre)1Cgn mice (see Stock No. 006054) and progeny lacking the neomycin cassette, but retaining loxP-flanked exon 1 were selected. Some of these mice were further backcrossed to C57BL/6J mice for at least 5 generations in the laboratory of Dr. Milena Bogunovic (University of Massachusetts Medical School). Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
