Detailed description

IL-21 KO mice contain a ZFN-mediated mutation targeted to exon 1 of the interleukin 21 (Il21) gene, creating a 1 bp deletion and early termination. Il21 encodes an immunoregulatory cytokine important in innate and adaptive immune responses. IL‐21 protein is not detected in stimulated splenocytes from IL-21 KO mice, and mice of both sexes are completely resistant to type 1 diabetes (T1D) development. They are also significantly protected from the development of lacrimal gland inflammation. Homozygous females and males are viable and fertile.

Development

This zinc finger nuclease induced mutation targets exon 1 of the interleukin 21 (Il21) gene, creating a 1 bp deletion and early termination. The mRNA encoding the ZFN was injected into one cell embryos derived from NOD/ShiLtJ mice (Stock No. 001976) and one mouse was identified by PCR and bred back to a NOD/ShiLtJ mice. Resulting offspring were bred to NOD/ShiLtJ mice to develop a colony of IL‐21 KO (line 39) mice. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize NOD/ShiLtJ oocytes.
