Pirnac1 (piRNA cluster 1; also called pi17) produces approximately 16% of all pachytene PIWI-interacting RNAs (piRNAs) in mouse pachytene spermatocytes. Unlike other pachytene piRNAs, loss of pi17/Pirnac1 has no detectable phenotype or impact on male fertility. These pi17em2 mice carry a CRISPR/Cas9-generated 517 bp knock-out deletion of the promoter and both transcriptional start sites of the two noncoding RNAs of the mouse Pirnac1 gene. Homozygous mice exhibit no overt phenotype, have normal body weight, are healthy, and are completely fertile. Homozygous mice are used in the study of pachytene piRNA clusters and male fertility. Males produce normal sperm counts and numbers of offspring.
