Description

Atoh1^FlpO knock-in/knock-out mice express codon-optimized Flp recombinase (FlpO) from the mouse Atoh1 (atonal bHLH transcription factor 1) promoter. Endogenous expression of Atoh1 is knocked out in this allele.

Detailed description

Atoh1^FlpO knock-in/knock-out mice express codon-optimized Flp recombinase (FlpO) from the mouse Atoh1 (atonal bHLH transcription factor 1) promoter. Expression of FlpO is found in cells that normally express Atoh1, including parabrachial complex neurons derived from the rostral rhombic lip (rRL). Endogenous expression of Atoh1 is knocked out in this allele.

Crosses with FlpO reporter strains enable tracing of Atoh1-lineage cells.

Heterozygous Atoh1^FlpO mice are viable and fertile. Homozygotes, lacking expression of Atoh1, are perinatal lethal.

Development

A FlpO-lox2722-PGK-Neo-lox2722 cassette was used to replace the coding sequence of the mouse Atoh1 gene, leaving the transcriptional start site intact. The mutation was created by homologous recombination in C57BL/6N-A^tm1Brd-derived JM8A3 embryonic stem (ES) cells. Resultant chimeric males were backcrossed to albino C57BL/6J female mice. Mice were subsequently crossed with Sox2^Cre mice on a C57BL/6 genetic background (Stock No. 008454) to excise the PGK-neo cassette. This strain was maintained on a C57BL/6 background by the donating laboratory (see SNP note below). The agouti and albino alleles were bred out of this strain; mice are black.

In 2021, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Four of the 43 markers throughout the genome, on Chromosomes 8, 12, and 19, were segregating with an unknown source. Also, one of the 5 markers that determine C57BL/6J from C57BL/6N was found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.
