Detailed description

Gata1^creERT2 BAC transgenic mice express erythroid lineage-specific, tamoxifen (Tx)-inducible cre recombinase from the mouse Gata1 (GATA binding protein 1) promoter in adult red blood cells.

Test crosses with R26 tdTomato (TdT) reporter mice (Stock No. 007914) were carried out to check for potential “leakiness” of Gata1^creERT2 in the absence of tamoxifen. Results demonstrated that fraction I (non-erythroid bone marrow) cells were almost completely TdT-negative, whereas >90% of the cells in fraction II (erythroid lineage-committed cells), were fluorescent. TdT intensity diminished during differentiation, possibly through a combination of chromatin condensation, TdT turnover and dilution. Very few TdT⁺ cells were present in the non-erythroid (B220/Gr1/CD11b/CD3ε⁺) population, indicating that Cre was active almost exclusively in the erythroid lineage.

Development

A tamoxifen-inducible CreER^T2 fusion gene was introduced to the start codon of the mouse Gata1 gene contained within BAC RP23-443E19 (also encoding genes Glod5, Hdac6, Eras, and Pqbp1). The modified BAC was introduced to C57BL/6J zygotes. The insertion site and copy number of the transgene are unknown. Expression of other genes located on the BAC (Glod5, Hdac6, Eras, and Pqbp1) has not been investigated.
