H19&Delta;13 knock-out mice have a neo cassette replacing the entire coding region of the H19, imprinted maternally expressed transcript (H19) gene. H19 is located in an imprinted region of chromosome 7 near the insulin-like growth factor 2 (Igf2) gene. H19 is only expressed from the maternally-inherited chromosome, whereas Igf2 is only expressed from the paternally-inherited chromosome. The product of this gene is a long non-coding RNA which functions as a tumor suppressor. Mutations in this gene have been associated with Beckwith-Wiedemann Syndrome and Wilms tumorigenesis. Defects are seen in heterozygotes that receive the mutant allele from the mother, while those that receive the mutant allele from the father are normal. When this mutation is maternally inherited, the maternal H19 gene is repressed and the normally imprinted Igf2 and Ins2 genes are expressed. These mice have a combined effect of the lack of H19 and the overexpression of IGF2. H19&Delta;13 knock-out mice are substantially larger than their WT littermates due to a double dose of Igf2. The mice are viable and fertile and besides the larger size do not show any other obvious phenotype. However, since homozygous offspring frequently cause dystocia in their mothers due to their large size, the donating investigator breeds heterozygous females with wildtype males only.
