Description

Foxl1CreERT2-2A-tdTomato knock-in/knock-out mice have an inducible Cre recombinase/ERT2 fusion followed by a tdTomato reporter inserted into the endogenous translation start site of the forkhead box L1 (Foxl1) gene. These mice can be to identify cells in the gastrointestinal tract.

Detailed description

Foxl1CreERT2-2A-tdTomato knock-in/knock-out mice carry an inducible Cre/ERT2 followed by a tdTomato reporter inserted into the endogenous translation start site of the forkhead box L1 (Foxl1) gene. The inserted sequences abolish expression of FOX1L. Foxl1 encodes a member of the forkhead/winged helix-box (FOX) family of transcription factors, it is expressed in the stomach and intestine. Mice that are hemizygous for this knock-in/knock-out allele are viable and fertile. Mice homozygous for Foxl1CreERT2-2A-tdTomato exhibit developmental delays in gut formation and abnormal proliferation and architecture of the gastrointestinal epithelium.

When crossed with a strain containing a loxP site flanked sequence of interest, the offspring are useful for generating induced cre-mediated targeted deletions.

Development

The Foxl1CreERT2-2A-tdTomato allele was created by Dr. Klaus Kaestner at the University of Pennsylvania. To create the strain a targeting vector containing a Cre recombinase/ERT2 fusion, a viral 2A oligopeptide that mediates ribosomal skipping, and a tdTomato reporter followed by neomycin resistance cassette was inserted after the ATG site in exon 1 of the Foxl1 gene on chromosome 8. The insertion deletes 200 bp of the Foxl coding sequence rendering the allele null for FOXL expression. The linearized construct was introduced by nucleofection into (C57BL/6 x 129S4/SvJae)F1-derived V6/5 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric animals were crossed to C57BL/6J females. Offspring were backcrossed to C57BL/6J for 6 generations. Upon arrival, mice were bred to C57BL/6J for at least 1 generation to establish the colony.
