Description

Gck^Cre mice express improved Cre recombinase (icre) from the mouse Gck (glucokinase) promoter, enabling the excision of loxP-flanked sequences in GCK-positive regions of the brain (e.g. in neurons of the paraventricular thalamus (PVT)). This strain has been useful in Cre-dependent optogenetic and chemogenetic studies of glucose sensing and feeding behavior.

Development

An FRT-flanked drug selection cassette was introduced to intron 8 and a T2A-iCre cassette was placed in exon 10 just before the Gck stop codon via homologous recombination in C57BL/6NTac-derived ART B6 3.6 embryonic stem (ES) cells. The drug selection cassette was excised with Flp recombinase. This strain was created and maintained on a C57BL/6N genetic background by the donating laboratory.

In 2022, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Four of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating, including the rd8 recessive mutation associated with retinal degeneration on Chromosome 1 within the Crb1 locus. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
