Exon 11 of the mouse Slc2a2 (solute carrier family 2 (facilitated glucose transporter), member 2; also called Glut2) gene is flanked by loxP sites in these Glut2^lox mice. Cre recombinase-mediated excision of the floxed region results in a presumptive knock-out allele.

Detailed description

Exon 11 of the mouse Slc2a2 (solute carrier family 2 (facilitated glucose transporter), member 2; also called Glut2) gene is flanked by loxP sites in these Glut2^lox mice. Exon 11 encodes the C-terminal 67 amino acids of the protein including the last transmembrane domain and the intracellular C-terminal tail. Western blot analysis confirmed normal protein expression from the floxed allele in the liver. Cre recombinase-mediated excision of exon 11 results in a presumptive knock-out allele.

Development

A loxP site was inserted in intron 10 and a loxP-FRT-PGK-neomycin-FRT cassette was placed in intron 12 via homologous recombination in 129P2/OlaHsd-Hprt^b-m3-derived HM-1 embryonic stem (ES) cells. Exon 11 encodes the C-terminal 67 amino acids of the protein including the last transmembrane domain and the intracellular C-terminal tail. This strain was backcrossed to C57BL/6J for an unknown number of generations by the donating laboratory. Assays conducted at The Jackson Laboratory demonstrate an absence of the neomycin cassette, thus it was likely excised with FLP recombinase.

In 2022, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Three of the 43 markers throughout the genome, on chromosomes 1, 3, and 12, were segregating between C57BL/6NJ, C57BL/6J, and 129, including the rd8 recessive mutation associated with retinal degeneration on Chromosome 1 within the Crb1 locus. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.
