Description

PAPP-A2 conditional mutant mice possess loxP sites flanking exon 2 of the Pappa2 (pappalysin 2) gene and may be useful in generating conditional mutations to study postnatal bone growth in mice.

Detailed description

Pappa2 (pappalysin 2 or pregnancy associated plasma protein A2) encodes a member of the pappalysin family of metzincin metalloproteinases and is involved in the regulation of insulin-like growth factor. These conditional Pappa2 mice possess loxP sites flanking exon 2. Mice homozygous for the mutation are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in cre-expressing tissues. Deletion of exon 2 is expected to result in the splicing of exon 1 to exon 3 creating a stop codon.

For example, when bred to B6.C-Tg(CMV-cre)1Cgn/J (Stock No. 006054), with widespread Cre recombinase expression, homozygous progeny have slight reduction in the weight and length of bones and cranium, as well as minor changes in skeletal microarchitecture, elevated circulating IGFBP5. These mice may be useful in generating conditional mutations to study postnatal bone growth in mice.

Development

The targeting construct contains from 5' to 3': a loxP site upstream of exon 2 and an FRT-flanked neomycin cassette followed by a second loxP site downstream of exon 2 of the Pappa2 gene. The construct was electroporated into B6.Cg-Thy1^a-derived Bruce4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Chimeric males were crossed to C57BL/6 females. Offspring were bred with B6.Cg-Tg(Pgk1-flpo)10Sykr/J (Stock No. 011065) mice to delete the neo cassette. The resulting progeny were crossed to remove the Flp-expressing transgene. The strain was maintained on a C57BL/6J background. Upon arrival, mice were bred to C57BL/6J (Stock No. 000664) for at least one generation to establish the colony.
