Description

Thy1.1 DO11.10 Balb/c mice carry the Thy1^a (Thy1.1) allelic variant and the Tg(DO11.10)10Dlo transgene, which contains rearranged Tcra and Tcrb genes encoding a chicken ovalbumin-specific MHC class II (I-A^d)-restricted T cell receptor. Thy1^a provides a second tracking option in adoptive transfer studies.

Detailed description

Thy1.1 DO11.10 Balb/c mice carry a Thy1^a (Thy1.1) allelic variant and a rearranged T cell receptor transgene, Tg(DO11.10)10Dlo, specific for the MHC class II (I-A^d)-restricted ovalbumin (OVA) peptide antigen. The Tg(DO11.10)10Dlo transgene is expressed in the majority of T cells and allows for examination of antigen-specific T cell responses.Mice homozygous for both alleles are viable and fertile. The addition of the Thy1^a allele to the DO11 transgene provides a second tracking option (in addition to the KJ126 antibody) for adoptive transfer studies and allows for more accurate tracking of transferred cells. These mice may be useful for a wide-range of studies of studies examining antigen-dependent T cell outcomes in vivo.

Of note, the DO11 transgene with the BALB-associated Thy1.2 (Thy1^b) allele is available as Stock No. 003303 C.Cg-Tg(DO11.10)10Dlo/J.

Development

The DO11 transgene was developed by Dr. Dennis Loh at Washington University School of Medicine. Constructs, about 45 to 50 kilobases, made from full genomic TCR rearranged genes from the DO11.10 hybridoma of Kappler and Marrack were co-microinjected into (B6C3F1) female X BALB male embryos. These rearranged transgenes encode a chicken ovalbumin-specific MHC class II (I-A^d)-restricted T cell receptor. The transgene integrated on distal chromosome 18. Mice from founder line 10 were bred back to BALB/c. The strain was transferred to The Jackson Laboratory as C.Cg-Tg(DO11.10)10Dlo/J (Stock No. 003303), Dr. Clara Abraham (Yale University) intercrossed it with mice carrying the Thy1^a (Thy1.1) variant on chromosome 9. This strain has been backcrossed at least 15 times to BALB/cJ (Stock No. 000651). Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize BALB/cJ oocytes (Stock No. 000651).

Expressed genes: Tcra,T cell receptor alpha chain, mouse, laboratory; Tcrb,T cell receptor beta chain, mouse, laboratory
Site of expression: T cells
Molecular note: This double transgene contains rearranged Tcra and Tcrb genes and is expressed in the majority of T cells. These rearranged transgenes from the DO11.10 T cell hybridoma encode a chicken ovalbumin-specific MHC class II (I-A^d)-restricted T cell receptor. The allele was created by injection of a mix of the two separate transgenes Tg(TcraDO11.10)#Dlo and Tg(TcrbDO11.10)#Dlo into oocytes.
General note:

Mice carrying this pair of MHC class II restricted rearranged Tcra and Tcrb T cell receptor transgenes on a H2^d background react to ovalbumin (OVA) peptide antigen. Intraperitoneal administration of OVA to transgenic mice results in a rapid deletion of the immature CD4⁺ CD8⁺ TCR^lo thymocytes with progression to mature thymocytes. Apoptosis of cortical thymocytes in these mice occurs within 20 hours of treatment.

General note:

The Thy1 locus determines a surface antigen present on cells of the thymus, a number of mouse leukemias, brain, and in lesser amounts on lymph node and spleen cells. The allele Thy1^a determines an antigenic specificity, Thy-1.1, found in the AKR and RF strains; the allele Thy1^b determines an antigenic specificity, Thy-1.2, found in the C3HeB/Fe and many other strains (J:5243, J:5012, J:4469). The Thy1 antigen is probably present on all T lymphocytes and absent from all B lymphocytes, and it thus serves as a valuable T-cell marker (J:5243). It is very widely used in experiments designed to determine the distribution and function of T-cells. Thy1 specifies a T-cell surface glycoprotein, T25, with a molecular weight of 25 kDa (J:5707). The protein appears to be anchored in the cell membrane by a lipid that is either phosphotidylinositol or closely related to it (J:12016). Thy1 may function in the cell membrane as a signal transduction molecule (J:8333). The Thy1 locus, or possibly a gene closely linked to it, controls quantitative expression of a protein that is the same size as Thy1 and is expressed on thymus and brain but not on lymph node and spleen cells (J:7900).
