Description

Klf2^GFP/GFP mice have an EGFP sequence inserted into the translational start site in exon 1 of the Klf2 gene, resulting in the expression of a KLF2-GFP fusion protein. These mice may be useful for imaging KLF2 expressing cells.

Detailed description

Klf2^GFP/GFP mice have an EGFP sequence inserted into the translational start site in exon 1 of the Kruppel-like factor 2 (lung) (Klf2) gene, resulting in the expression of a KLF2-GFP fusion protein. Klf2 encodes a zinc finger transcription factor expressed early in mammalian development and may be involved in adipogenesis, embryonic erythropoiesis, cardiovascular development, endothelial physiology, inflammation and T-cell trafficking and function.

Klf2^GFP/GFP mice have normal numbers of CD4+ and CD8 T+ cells. These mice recapitulate endogenous KLF2 expression, as EGFP is not expressed until the mature stage of single positive (SP) thymocyte development. KLF2 is downregulated during T cell activation but is upregulated again in some T cell populations. Fluorescence is detectable by flow cytometry or immunohistochemistry.

Mice that are homozygous for this allele are viable and fertile.

Development

The Klf2^GFP targeting vector was designed to insert an enhanced green fluorescent protein (EGFP) sequence into the translational start site in exon 1 of the Kruppel-like factor 2 (lung) (Klf2) gene. A loxP-flanked neomycin resistance (neo) cassette was inserted downstream of exon 2. The construct was electroporated into C57BL/6-derived embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts. The resulting Klf2^GFP mice were bred to CMV-cre mice (Stock No. 006054) to remove the neo cassette, and resulting mice were subsequently bred to C57BL/6NCrl mice for at least 4 generations by the donating laboratory (see SNP note below). Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

In 2021, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Two of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
