Description

TIPF floxed mice have loxP sites flanking exons 2-4 of the Tax1bp3 gene. These mice are useful for studying development and inhibition of the Wnt signaling pathway.

Detailed description

TIPF floxed mice have loxP sites flanking exons 2-4 of the Tax1 (human T cell leukemia virus type I) binding protein 3 (Tax1bp3) gene. Tax1bp3 encodes a protein that interacts with a large number of target proteins through protein-protein interactions with its PDZ domain. It plays a role in cell signaling, adhesion, protein scaffolding, and receptor and ion transporter functions and may act as an inhibitor of the Wnt signaling pathway. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 2-4 deleted in the cre-expressing tissues.

Development

The L1L2_Bact_P cassette was inserted at a position on Chromosome 11 upstream of exon 2 of the Tax1 (human T cell leukemia virus type I) binding protein 3 (Tax1bp3) gene. The cassette is composed of an FRT site followed by a lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance (neo) gene under the control of the human β-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of exon 4. The mutation was generated in C57BL/6N-A^tm1Brd-derived JM8A1.N3 embryonic stem (ES) cells, and correctly targeted ES cells of clone HEPD0576_5_A06 were injected into blastocysts. The resulting chimeric mice were bred to B6 ROSA26Flpo mice (Stock No. 012930) to remove the FRT-flanked lacZ sequence and neo cassette. Resulting TIPF floxed mice were bred to C57BL/6NCrl mice for at least 6 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Molecular note: The L1L2_Bact_P cassette was inserted at a position on Chromosome 11 upstream of exon 2. The cassette is composed of a FRT site followed by a lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance (neo) gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of exon 4. Flp-mediated recombination removed the FRT-flanked neo cassette and lacZ reporter.
