Description

LanCL3 KO mice carry a knock-out allele of the X-linked mouse Lancl3 (LanC lantibiotic synthetase component C-like 3 (bacterial)) gene in which most of exon 1 and exon 2-5 were removed by a 37 bp deletion. This strain has been useful in studies of LanCL protein function.

Detailed description

LanC-like (LanCL) enzymes respond to sites of protein damage. Lancl3 (LanC lantibiotic synthetase component C-like 3 (bacterial)) is normally expressed in a variety of tissues, with the most abundant expression in the cerebral cortex.

These LanCL3 KO mice carry a knock-out allele of the X-linked mouse Lancl3 gene in which most of exon 1 and exons 2-5 were removed by a 37 bp deletion. Absence of the protein was confirmed by immunoblot analysis (He, et al. 2017).

No distinct phenotype has been associated with LanCL3 KO mice, but this strain has been useful in studies of LanCL protein function. Triple knock-out mice incorporating this LanCL3 KO allele (Stock 036163) in addition to Lancl1 (Stock No. 029694) and Lancl2 (Stock No. 029695) knock-out alleles may be at risk of premature death, but no underlying cause has been identified (Lai, et al. 2021).

Development

Zinc finger nuclease (ZFN) mRNA designed to target the third exon of the X-linked mouse Lancl3 gene was injected into FVB embryos. This created a 37 bp deletion incorporating most of exon 1 and exons 2-5. Resultant mice were backcrossed to C57BL/6N for 10 generations by the donating laboratory (see SNP note below). Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

In 2022, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Three of the 43 markers throughout the genome were segregating with FVB. Also, one of the 5 markers that determine C57BL/6J from C57BL/6N was found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.

Molecular note: Pronuclear-stage embryos were targeted with a zinc finger nuclease pair designed to target exon 1. A founder mouse was identified carrying a 37 nucleotide deletion at the targeted site. Absence of the protein was confirmed by immunoblot analysis.
