Description

Esr1^flp knock-in mice express optimized FlpO recombinase from the mouse Esr1 (estrogen receptor 1 (alpha)) promoter. This enables the excision of FRT-flanked sequences of interest in estrogen receptor 1-expressing cells, including neurons of the medial preoptic area (MPOA) and ventromedial hypothalmus, ventrolateral subdivision (VMHvl).

Detailed description

Esr1^flp knock-in mice express optimized FlpO recombinase from the mouse Esr1 (estrogen receptor 1 (alpha)) promoter. This enables the excision of FRT-flanked sequences of interest in estrogen receptor 1-expressing cells, including neurons of the medial preoptic area (MPOA) and ventromedial hypothalmus, ventrolateral subdivision (VMHvl). As FlpO was introduced to the 3' end of the Esr1 coding sequence, Esr1 expression is not anticipated to be disrupted.

Crosses with mice expressing Slc32a1 (Vgat)^cre (Stock No. 028862) have enabled researchers to manipulate subpopulations of inhibitory Esr1-expressing neurons via optogenetic activation in behavioral studies (Pubmed: 33268894).

Development

An attP-F2A-Flp-lox2272-Pgk-Neo-pA-lox2272-attP cassette was introduced to the 3' UTR of the mouse Esr1 gene by homologous recombination in 129S6/SvEvTac-derived TC-1 embryonic stem (ES) cells. Resultant male mice were bred with C57BL/6N female mice. The neomycin cassette was excised. This line was subsequently backcrossed to C57BL/6NCrl for more than 14 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

In 2021, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. One of the 5 markers that determine C57BL/6J from C57BL/6N was found to be segregating on Chromosome 6. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed C57BL/6N;C57BL/6J genetic background.
