Description

PYY-GFP mice express an enhanced green fluorescent protein (EGFP) under the control of the Pyy (peptide YY) promoter. The PYY peptide hormone is produced by the L cells of the gastrointestinal mucosa and functions as a major signal of satiety. This mutant mouse strain may be useful in visualization and FACS isolation of PYY secreting cells.

Detailed description

Pyy (peptide YY or peptide tyrosine tyrosine) encodes a peptide hormone that is produced and secreted by enteroendocrine cells of the small intestine and colon.  PYY also is secreted by a subset of taste buds. PYY functions to induce satiety and thus reduce weight gain. PYY is also involved in the regulation of taste, esophageal muscle contraction, gastric emptying, colonic mobility, vasoconstriction, and sodium excretion. The PYY-GFP transgene uses the mouse Pyy (peptide YY) regulatory elements to drive expression of the enhanced green fluorescent protein (EGFP) in PYY hormone producing cells. EGFP expression is observed in the pyloric region of the gastric mucosa to the distal descending colon, however, the majority of PYY-GFP cells are found in the distal ileum and colon. EGFP expressing cells are immunoreactive with a PYY antibody and can also be observed using endogenous green fluorescence. Mice hemizygous and homozygous for the transgene are viable and fertile. This mutant mouse strain may be useful in visualization and FACS isolation of PYY secreting cells.

Development

The transgenic construct contains 895 bp of sequence (A box) from directly upstream of the mouse Pyy start codon and 681 bp of sequence (B box) from downstream of the mouse Pyy stop codon obtained from bacterial artificial chromosome RP23-218N17. An EGFP-BGH sequence followed by an FRT-flanked neomycin/kanamycin cassette was inserted between the Pyy A and B boxes. The neomycin/kanamycin cassette was removed by arabinose induction of flipase in bacterial cells. The BAC was linearized and microinjected into the pronuclei of fertilized B6SJLF1 oocytes. Founder line 2 was established. The colony was maintained on a C57BL/6J and SJL background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
