Description

Col2a1-tTA transgenic mice express tTA (tetracycline-controlled transactivator) from the chondrocyte-specific mouse Col2a1 (collagen, type II, alpha 1) promoter/enhancer. When combined with a tetO/TRE transgene, expression of the target gene of interest can be blocked in chondrocytes by administration of doxycycline in a "Tet-Off" system.

Detailed description

Col2a1-tTA transgenic mice express tTA (tetracycline-controlled transactivator) from the chondrocyte-specific mouse Col2a1 (collagen, type II, alpha 1) promoter/enhancer. When combined with a tetO/TRE transgene, expression of the target gene of interest can be blocked in chondrocytes by administration of doxycycline in a "Tet-Off" system.

In combination with the TRE-H2B-EGFP allele (Col1a1^{tm7(tetO-HIST1H2BJ/GFP)Jae}, which may be isolated from Stock No. 016836), these Col2a1-tTA mice enable a genetic approach to fluorescently isolating slow-cycling label-retaining chondrocytes (LRCs) from the growth plate of bone based on a chondrocyte-specific, doxycycline-driven pulse-chase protocol.

Development

An 8.4kb construct incorporating the mouse Col2a1 promoter and a fragment of intron 1 ligated to a splice acceptor sequence followed by an internal ribosome‐entry site (IRES), tetracycline‐controlled transactivator (tTA), and the SV40 large T antigen polyadenylation signal was introduced to B6SJLF1 fertilized eggs. A single copy of the Col2a1‐tTA transgene was inserted 16 kb downstream of Peli2 (pellino 2) on mouse Chromosome 14. This strain was backcrossed to FVB/N for at least for 8 generations by the donating laboratory. This is founder Line H.
