The Cebpa Enh/prom-hCD4 transgene was designed with the hematopoietic-specific CCAAT/enhancer binding protein (C/EBP), alpha (Cebpa) + 37 kb enhancer and Cebpa promoter directing expression of truncated human CD4 antigen (CD4) cDNA. Cebpa is an intronless gene that encodes a basic region-leucine zipper transcription factor involved in proliferation arrest and the differentiation of myeloid progenitors, adipocytes, hepatocytes, and cells of the lung and the placenta. Cebpa contains an evolutionarily conserved 440 bp enhancer +37 kb upstream of the ATG site, that directs high-level expression to myeloid progenitors. This enhancer has 85% homology to the +42 kb region of the human CEBPA locus, harboring enhancer specific histone marks, and together with the promoter is capable of directing high-level hCD4 transgene expression to granulocyte-monocyte progenitor (GMP), common myeloid progenitor (CMP), and Lin-Sca-1+c-kit+ (LSK) stem/progenitor cell subsets, and moderate expression to a subset of common lymphoid progenitors (CLP), but not to multiple non-hematopoietic tissues. Cytoplasmic truncation of hCD4 prevents signals from being sent into the cell, and its expression at the plasma membrane allows detection by FACS antibodies that do not cross-react with murine CD4. Hemizygous mice are viable and fertile. The donating investigator has not attempted to make a homozygous colony and reports that hemizygous mice are smaller when young and mature slower than wildtype mice. These mice are useful for studies investigating mechanisms that underlie hematopoietic lineage specification.

Typically mice are recovered in 12-16 weeks. Contact Customer Service to place an order or for more information.
