Tim-3 Tg mice have a human CD2 antigen (CD2) promoter directing expression of full-length BALB/c isoform of hepatitis A virus cellular receptor 2 (Havcr2) cDNA. Havcr2, also known as Tim-3, is a Th1-specific cell surface protein that regulates macrophage activation, inhibits Th1-mediated auto- and alloimmune responses, and promotes immunological tolerance. In the thymus, the Tim-3 Tg is expressed at the double negative stage and maintained through the double positive and single positive (SP) stages of thymocyte development. Tim-3 Tg is expressed in only 30&ndash;40% of CD4+SP and CD8+SP thymocytes in the thymus and the periphery. Analysis of thymic development showed a small decrease in the frequency of Tim-3 Tg+ double negative thymocytes, but no significant difference in the frequency of Tim-3 Tg+ double positive thymocytes. These mice exhibit depressed T cell responses in comparison with wildtype control littermates and they contain a higher frequency of CD11b+Ly-6G+ myeloid suppressor cells. Hemizygous mice are viable and fertile, the donating investigator has not attempted to make a homozygous colony.
