Description

Dmrt3 (doublesex and mab-3 related transcription factor 3) knock-out mice exhibit a reduction in size of the cerebral hemispheres and the hippocampus. These mice may be useful for studies involving the regulation of patterning of the cerebral cortex.

Detailed description

Dmrt3 (doublesex and mab-3 related transcription factor 3) encodes a DNA-binding transcription factor expressed in cortical progenitors in a high caudomedial to low rostrolateral gradient and is involved in cortical patterning. Dmrt3 knock-out mice contain a neomycin cassette that replaces exon 1, which contains the DM domain. Mice homozygous for the mutation are viable, but are not used for breeding due to tooth overgrowth and behavioral issues. Homozygous Dmrt3 knockout mice exhibit decreased size of cerebral hemispheres and the hippocampus, tooth overgrowth, runting, and behavioral issues. This strain may be useful for studies involving the regulation of patterning of the cerebral cortex.

Development

The targeting construct was designed to replace exon 1 of the Dmrt3 gene with a loxP-flanked neomycin cassette. Exon 1 includes the DM domain, a zinc finger-like DNA-binding motif, as well as the transcription start site. The construct was electroporated into 129S1/Sv-derived CJ7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Chimeric males were crossed to C57BL/6J females. Offspring were bred with mice carrying Tmem163^{Tg(ACTB-cre)2Mrt} on a congenic C57BL/6 background to delete the neo cassette. The resulting progeny were crossed to remove the cre-expressing transgene. The colony was crossed to C57BL/6J for approximately 7 generations by the donating lab (see SNP notes below). Upon arrival, mice were bred to C57BL/6J (000664) for at least one generation to establish the colony.

In 2021, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Most of the 43 markers throughout the genome were segregating, suggesting an incomplete backcross.

Symbol: Dmrt3^tm1.1Zark
Name: targeted mutation 1.1, David Zarkower
MGI accession ID: MGI:6471086
Generation method: Targeted
Attributes: Null/Knockout
Marker symbol: Dmrt3
Marker name: doublesex and mab-3 related transcription factor 3
Marker synonyms: DMRTA3
Chromosome: 19
Strain of origin: 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺
Molecular note: The targeting construct is designed to replace exon 1 with a loxP-flanked neomycin cassette. Exon 1 includes the DM domain, a zinc finger-like DNA-binding motif, as well as the transcription start site. Cre-mediated recombination removed the floxed neo cassette.
