Description

SAA^3KO (SAA3 KO) CRISPR/Cas9-generated knock-in/knock-out mice carry premature stop codons and a frame shift mutation in exon 2 of the mouse Saa3 (serum amyloid A 3) gene. This strain has been useful in studies of Th17 cell responses, inflammation, and autoimmunity.

Detailed description

Saa3 (serum amyloid A 3) plays a role in directing pathogenic pro-inflammatory Th17 cell differentiation, and is induced in the CNS during experimental autoimmune encephalomyelitis (EAE). SAA^3KO (SAA3 KO) CRISPR/Cas9-generated knock-in/knock-out mice carry premature stop codons and a frame shift mutation in exon 2 of the mouse Saa3 gene. SAA3 translation is terminated prematurely.

Development

Premature stop codons (TAG TAA), a frame shift mutation (A), and a HindIII site (AAGCTT) were introduced to exon 2 of the mouse Saa3 gene by CRISPR/Cas9 homology directed repair methodology. Nucleotide sequence cCTGGGAgttgacagccaaagatg became cTAGTAAAAAGCTTttgacagccaaagatg via the gRNA. This mutation was created using C57BL/6J embryos and backcrossed 6 generations further to C57BL/6J by the donating laboratory.
