Serum amyloid A proteins (SAAs) dictate the balance between homeostatic and inflammatory Th17 cells. LoxP-STOP-loxP-SAA1 (also called LSL-SAA1) knock-in mice carry a conditional mutant allele in which Saa1 (serum amyloid A 1) and IRES-EGFP are over-expressed from a CAG promoter upon cre recombinase-mediated excision of a loxP-flanked neomycin/Stop cassette. The functionality of the EGFP reporter has not been assessed. When crossed with Alb-cre mice (Stock No. <a href="https://www.jax.org/strain/003574" target="_blank">003574</a>), liverSAA1 progeny exhibit high concentrations of SAA1 in serum, due to constitutive production in liver. Numbers of Th17 cells, but not Th1 cells, are increased in secondary lymphoid tissues of liverSAA1 mice. Following myelin oligodendrocyte glycoprotein (MOG) immunization with a low dose of the attenuated adjuvant Mycobacterium tuberculosis (H37RA) antigen emulsified in incomplete Freund's adjuvant (IFA), liverSAA1 mice have increased experimental autoimmune encephalomyelitis (EAE) disease incidence compared to control littermates, suggesting that systemic elevation of SAA1 in liverSAA1 mice potentiates pathogenic Th17 effector function.
