Description

Irp2^flx mutant mice possess loxP sites flanking exon 3 of the Ireb2 (iron responsive element binding protein 2 or Irp2) gene and may be useful in generating conditional mutations to study iron distribution and hematopoiesis.

Detailed description

Ireb2 (iron responsive element binding protein 2) encodes an RNA-binding factor involved in the regulation of iron levels through the binding of iron-response elements (IRES). Conditional Irp2^flx mice possess loxP sites flanking exon 3 of the Ireb2 (or IRP2) gene. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have the exon 3 deleted in cre-expressing tissues.

For example, when Irp2^flx mice are bred to B6.C-Tg(Pgk1-cre)1Lni/CrsJ (Stock No. 020811) with widespread germline Cre recombinase expression, progeny exhibit microcytosis, some erythropenia, altered body iron distribution (iron loading in liver and duodenum) and iron deficiency in the spleen

Development

The targeting vector, created in the laboratory of Dr. Matthias Hentze (EMBL), contains a loxP site and an FRT-flanked IRES- βGeo cassette upstream of exon 3 and a loxP site downstream of exon 3 of the Ireb2 gene on chromosome 9. The construct was electroporated into 129P2/OlaHsd -derived E14 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Chimeric males were crossed to C57BL/6J females. Offspring, were bred with B6.129S4-Gt(ROSA)26Sor^tm1(FLP1)Dym/RainJ mice to delete the βGeo cassette. The resulting progeny were crossed to remove the flp-expressing transgene. Dr. Mark Fleming (Children's Hospital Boston) obtained Irp2^flx mice from Dr. Hentze and introgressed the allele into C57BL/6J for 10 generations. Upon arrival, mice were bred to C57BL/6J for at least 1 generation to establish the colony.
