Detailed description

Mmp3 null mice carry a CRISPR/Cas9 generated 1 bp deletion in the catalytic domain of the matrix metallopeptidase 3 (Mmp3), resulting in a premature stop codon. MMP family members are involved in the breakdown of extracellular matrix in normal physiological processes. MMP3, or stromelysin-1, is an enzyme which degrades fibronectin, laminin, collagens III, IV, IX, and X, and cartilage proteoglycans, and is thought to be involved in wound repair, progression of atherosclerosis, and tumor initiation.

Homozygous mice are viable and fertile. Created on a coisogenic C57BL/6J background, these mice should lack any potential 129-derived passenger mutations as seen in other mutant strains containing 129 embryonic stem cell-derived passenger flanking DNA (PMID: 26163370).

Development

A guide RNA was designed to delete one base pair (T) in exon 4 (Ensembl exon ID# ENSMUSE00000215695), on Chromosome 9 position 7,447,544 bp, in the catalytic domain of the matrix metallopeptidase 3 (Mmp3) gene. This mutation is predicted to cause amino acid sequence changes after residue 176 and a premature stop codon at position 327. sgRNA (GAGACTTTGTCCCTTTTGA) and the cas9 nuclease were introduced into the cytoplasm of C57BL/6J-derived zygotes with well recognized pronuclei. Correctly targeted embryos were transferred to pseudopregnant females. Mmp3 null pups were identified by PCR and sequencing, and founder mice were further bred to C57BL/6J mice for at least 4 generations to develop the colony. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J (Stock No. 000664) oocytes.
